PLACENTA – MOVIE 2: Human placenta, w19, vessels and cytotrophoblast layer in 3D.
The placenta is a large organ in the woman’s uterus during pregnancy, which provides oxygen and nutrients to the embryo/foetus and removes waste products from the baby’s blood. These videos reveal some details about the fine structure of human placenta in three-dimension, using the cytotrophoblast marker PEG10 as well as the immunomarker CD34, which depicts the blood vessels inside the organ.
METHOD: iDISCO+ volume immunostaining and clearing
TISSUE: Human placenta from abortion, w19.
ANTIBODIES: PEG10 (green); CD34 (red)
MOVIE DESCRIPTION: Same scan as in ‘Placenta MOVIE 1’ but now in 3D. The vessels and the cytotrophoblast layer are morphologically intact. The CD34 channel has a gradual decrease in signal intensity: this is a technical issue, due to limited laser penetration through this compact structure.
